Avocado Soy Unsaponifiables (ASU) are natural vegetable extracts made from avocado and soybean oils unsaponifiable fractions, which are the oily substances that remain after hydrolysis (saponification) of avocado and soybean oils. The major components of ASU are phytosterols such as beta-sitosterol, campesterol, and stigmasterol, which are considered anti-inflammatory compounds with both antioxidant and analgesic activities.
Phytosterols (plant sterols) from ASU preparations are thought to be the primary sources of protective activity for cartilage-producing cells. ASU reduces damage to cartilage by increasing the synthesis of collagen and proteoglycan (aggrecan), and preserves hydroxyproline, the component in collagen that makes it stable. It inhibits MMP3 and other pro-inflammatory signaling molecules.
A study conducted on patients with osteoarthritis of the hip not only found symptomatic improvement, but also determined ASU reduced the rate of progression of arthritis in the joints.
ASU blocks pro-inflammatory chemicals, prevents deterioration of synovial cells, which line joints, and may help regenerate normal connective tissue.

Mechanism of Action:

The cartilage between our joints becomes more susceptible to degradation with time, leading to stiffness, discomfort and, in some cases, osteoarthritis (OA).

Collagen is the main structural component of the various connective tissues in humans, including tendons, ligaments, cartilage and skin. Type II collagen makes up 50% of all protein in cartilage and have been studied in relation to joint health.
UC-II® has a unique mode of action that has been shown to initiate cell-signaling cascades that use the body’s natural repair mechanisms in the joints — and therefore plays a role in supporting the rebuilding of cartilage. But how exactly does UC-II undenatured type II collagen support the mobility of knee joints?
When taken orally, it is hypothesized that the intact type II collagen reaches specialized lymphoid follicles found in portions of the small intestine. These areas are known as Peyer’s Patches and form a part of the gut-associated lymphoid tissue.
The Mechanism of Action shows that, once the UC-II undenatured type II collagen makes contact with the Peyer’s Patches, it starts interacting with the immune system. The interaction is with a special class of cell, called a regulatory T-cell, which goes on to produce several regulatory molecules that are transported by the body to the joint.
Upon arrival at the joint, regulatory molecules, called cytokines, induce knee joint resident chondrocytes to begin producing more type II collagen and other constituents that make up the structural matrix of the knee joint.

ANTI-INFLAMMATORY JOINT RELIEF BLEND
Turmeric Root Extract:
Curcumin is one of the most studied natural molecules for its anti-inflammatory properties, it is a broad-spectrum ingredient and is the chemical in turmeric that can reduce joint pain and swelling by blocking inflammatory cytokines and enzymes. A 2010 clinical trial using a turmeric supplement showed long-term improvement in pain and function in patients with knee OA.
Curcumin helps reduce pain, inflammation and stiffness related to rheumatoid arthritis (RA) and osteoarthritis (OA) & treats bursitis. A potent anti-inflammatory, curcumin blocks inflammatory cytokines and enzymes, including 5-LOX and cyclooxygenase-2 (COX-2), the target of the drug celecoxib.
Ginger Root Powder:
Ginger has been shown to have anti-inflammatory properties similar to ibuprofen, the COX-2 inhibitor celecoxib and even TNF blockers. The anti-inflammatory properties seem to help relieve pain and improve function for all types of arthritis. The compounds in ginger can function as a COX-2 inhibitor, the same way as common medications for osteoarthritis. One review of 5 studies found that those with osteoarthritis who took ginger had a 30% decrease in pain and a 22% reduction in disability, compared with control groups.
A University of Miami study concluded that ginger extract could one day be a substitute to nonsteroidal anti-inflammatory drugs (NSAIDs). The study compared the effects of a highly concentrated ginger extract to placebo in 247 patients with osteoarthritis (OA) of the knee. The ginger reduced pain and stiffness in knee joints by 40 percent over the placebo.
White Willow bark:
The salicylic acid in white willow bark lowers the body’s levels of prostaglandins, hormonelike compounds that can cause aches, pain, and inflammation. While white willow bark takes longer to begin acting than aspirin, its effect may last longer. And, unlike aspirin, it doesn’t cause stomach bleeding or other known adverse effects.
Some arthritis sufferers taking white willow bark have experienced reduced swelling and inflammation, and increased mobility, in the back, knees, hips, and other joints.
White Willow bark contains an ingredient called salicin, which your body makes into another chemical substance called salicylic acid. Similar to acetylsalicylic, salicylic acid reduces the production of certain prostaglandins (hormone-like substances that control your immune system and fight joint inflammation) in your nerves, and this eases pain and discomfort. Willow bark showed anti-inflammatory activity in several laboratory-based studies.
Vitamin C, chemically identified as ascorbic acid, is a well-established anti-oxidant with biological proven tissue-based impacts, such as playing an essential role in collagen formation, one of the main components of cartilage, as well as having broader neurological effects crucial to the maintenance of the skeleton and possibly to its associated tissues and body systems. Moreover, its ability to foster multiple enzymatic processes and decrease inflammation, while influencing bone metabolism favorably is likely to be more helpful than not in preventing or attenuating osteoarthritis disease processes and their negative painful and disabling consequences.
Other research shows vitamin C, an essential cofactor for fostering collagen linkage production, ligament, tendon and bone quality is often present at marginal levels in people with arthritis, especially, if multiple drugs including painkillers and anti-inflammatory drugs are being used.
A further body of research implies that the presence of sufficient levels of vitamin C may also promote tendon and muscle collagen synthesis, while improving the antioxidative capacity of the synovial fluid, all possible structural changes that could lower the degree of any existing osteoarthritis pain while decelerating the disease progression.
The active components (Boswellic acids) are powerful anti-inflammatories with analgesic (pain-relieving) properties. It also may help prevent cartilage loss and inhibit the autoimmune process. In a 2008 study, the extract, also known as Loxin 5, significantly improved OA pain and function within seven days. An Indian study also revealed it slowed cartilage damage. Boswellia has been shown to lower the enzyme that stimulates the production of inflammation-causing mediators in the body. Clinical studies have shown that boswellia extracts improve pain symptoms more than a placebo in people with osteoarthritis.
Conclusion: Boswellia serrata extract reduced pain and improved physical functioning significantly in osteoarthritis patients; and it is safe for human consumption. 5-Loxin may exert its beneficial effects by controlling inflammatory responses through reducing proinflammatory modulators, and it may improve joint health by reducing the enzymatic degradation of cartilage in osteoarthritis patients.
Abstract: Osteoarthritis is the most common degenerative joint disease that begins asymptomatically in middle age with progressive symptoms in advancing age. Majority of people by the age 40 years may develop osteoarthritis, especially in weight bearing joints. Females are prone with 25% prevalence, whereas males have a prevalence of 16%. In the present study, 56 patients fulfilling the diagnostic criteria of osteoarthritis, divided into two groups. After a course of therapy for 2 months, symptomatic improvement was observed in both the groups at various levels with promising results in the patients of first group.
